High Expression of Actin Binding Proteins Predicts Hematogenous Metastases in Non-Small Cell Lung Cancer

Abstract

Introduction: The high mortality rate in non-small cell lung cancer (NSCLC) patients is associated with tumor progression. Tumor cell motility, which is mediated by various actin-binding proteins (ABPs) plays a crucial role in tumor progression. Therefore, the study of the relationship between the expression of ABPs (CAP1, cofilin-1, profilin-1, fascin-1 and ezrin) and NSCLC progression is of great significance. Methods: NSCLC tissue samples and tumor-adjacent unchanged lung tissue samples were collected from 46 NSCLC patients who were not previously treated. The mRNA expression of ABPs was determined by real-time PCR, and the ABP expression was analyzed using Western blotting assay. Results: All ABPs showed the correlation with 2-year metastasis-free survival. The increased mRNA levels of САР1, CFL1, EZR, FSCN1 and PFN1 and their protein products in tumor tissue compared to tumor-adjacent unchanged tissue indicated the risk of hematogenous metastases. Conclusion: The data obtained demonstrated the feasibility of using these ABPs as additional predictors of  NSCLC prognosis.
